Summary

TPD52 encodes tumor protein D52, and DGAT1 encodes diacylglycerol O-acyltransferase 1. A variant that affects both TPD52 and DGAT1 may play a role in rheumatoid arthritis by altering the response to immunosuppressant drugs and impairing sugar and fat metabolism. Lifestyle, diet, and supplement modifications may counteract the effects of these variants by correcting metabolism.

TPD52 and Rheumatoid Arthritis

The TPD52 gene encodes tumor protein D52. We know very little about TPD52 except that it can be used as a biomarker for some forms of cancer and that it has been associated with rheumatoid arthritis [R, R, R, R].

In cancer, lower levels of TPD52 are generally associated with worse outcomes [R].

Variations in TPD52 may affect whether or not certain rheumatoid arthritis treatments work well. In one study, researchers identified TPD52 as one of the rheumatoid arthritis susceptibility genes that might affect response to infliximab, a drug that suppresses the immune system [R, R].

Another Possible Connection: DGAT1

The TPD52 variant that has been associated with rheumatoid arthritis, rs998731, may also affect the expression of another gene called DGAT1. The DGAT1 gene encodes diacylglycerol O-acyltransferase 1 [R, R].

DGAT1 affects the storage and metabolism of sugar and fat. Recently, some studies have found that rheumatoid arthritis patients are less likely to be able to process sugars and fats in a healthy way compared with healthy people [R].

A genetic variant that may affect both TPD52 and DGAT1 has been associated with rheumatoid arthritis. TPD52 is best known as a cancer biomarker, while DGAT1 affects sugar and fat metabolism.

Aquatic Exercise

Variants of TPD52 and DGAT1 may alter sugar and fat metabolism in a harmful way. One study found that 45 minutes of hard exercise increased energy expenditure well after the physical activity was over. This revved up metabolism lasted for 14 hours [R, R].

Exercise can increase muscle mass, which helps boost metabolism, even at rest [R, R, R].

Aquatic exercise is often recommended to patients with rheumatoid arthritis. Aquatic exercise describes any type of physical activity that takes place either partially or fully submerged in water. These activities can allow people to maintain physical fitness and reap the benefits of exercise even when they are not fully mobile out of the water [R].

In 2 clinical trials of over 250 people with rheumatoid arthritis comparing the effects of different types of exercise, aquatic exercise produced the greatest pain reduction [R, R].

Aquatic exercise may help correct problems with metabolism and improve the symptoms of rheumatoid arthritis.

Fish Oil

Fish oil’s anti-inflammatory effects have the ability to indirectly aid in fat metabolism in people with high inflammation [R].

It can increase the secretion of adiponectin, which is responsible for breaking down fats [R].

Supplementation with fish oil may lower the activity of chronic inflammatory and autoimmune diseases, including rheumatoid arthritis [R, R].

A meta-analysis of 17 trials found that supplemental fish oil reduced inflammatory joint pain from rheumatoid arthritis and other inflammatory conditions [R].

In one study of 250 patients, fish oil was as effective as ibuprofen in reducing pain caused by arthritis [R].

Resolvins found in EPA and DHA appear to prevent certain inflammatory cytokines such as TNF-α from inducing pain [R].

Fish oil may help improve fat metabolism and reduce inflammation and pain in rheumatoid arthritis.

Vitamin E

People with impaired fat metabolism, which may be caused by DGAT1 changes, are more likely to be deficient in vitamin E [R, R, R, R].

One study found that vitamin E supplements (600 mg twice a day for twelve weeks), when added to conventional rheumatoid arthritis therapy, improved pain but not inflammation [R].

In multiple studies of animals with rheumatoid arthritis, vitamin E slowed the progression of joint destruction [R, R, R].

People with DGAT1 variants may have impaired fat metabolism and may therefore be more susceptible to vitamin E deficiency.
