The PTHLH gene codes for parathyroid hormone-like hormone (PTHLH), a protein that suppresses the growth and expansion (proliferation) of cells. In hair follicles, the production of PTHLH must be tightly regulated in order to maintain proper hair growth cycles [R, R, R].
Hair growth cycles include cell proliferation (hair growing), follicle regression (hair detaching), and rest (hair falling out). In line with its ability to suppress cell proliferation, PTHLH levels have been observed to steadily rise over the course of hair growth, and reach their peak prior to hair detachment [R, R].
Studies have shown that this effect may be due to prevention of blood vessel formation by PTHLH. Blood vessels provide hair follicle cells with oxygen, nutrients, and other factors essential for their growth [R, R].
Interestingly, PTHLH may also be necessary for transitioning follicles from rest to proliferation, suggesting a general role for this hormone in stimulating and accelerating the hair growth cycle. The transition from rest to proliferation may be dependent on proteins beta-catenin and LEF-1 (part of the Wnt signaling pathway) [R, R, R].
Variants of PTHLH have been associated with male-pattern baldness. These variants may dysregulate the production of PTHLH, resulting in lower beta-catenin and LEF-1 levels and hair cycle perturbation [R, R, R, R].
