Mechanism:

  • Minor “T” allele= increased SCN2A production, which contributes to increased neuronal firing throughout the brain [R].

The T (minor) allele is associated with:

  • Higher general cognitive ability [R].
  • If diagnosed with schizophrenia the minor “T” allele is associated with worse cognitive function [R].
  • This genetic advantage may be mediated by increased capacity for information processing between the dorsolateral prefrontal cortex and dorsal anterior cingulate cortex, which support higher cognitive functions [R].
  • In schizophrenia, the minor T allele was associated with relatively decreased SCN2A production in dorsolateral prefrontal cortex (dlPFC). It was also associated with relatively increased or inefficient dlPFC activity during a working memory task [R].

Summary

T (minor allele)=significantly higher general cognitive or “g” independent of gender and age (R).

This single SNP explained approximately 2–5% of the variance in “g” with higher scores in carriers of the minor T allele (R), which is quite a lot for a single SNP.

The  SCN2A  gene encodes the α2 subunit of voltage-gated type II sodium channels, which contribute to neuronal firing throughout the brain (R).

Convergent evidence across three independent cohorts of healthy adults (n=531) demonstrate that the T allele predicts human “g.” (R)

However, in schizophrenia, the T allele is associated with worse cognitive function (R).
